撫でる (naderu) is a general, gentle stroking motion, often in one direction. さする (sasuru) implies a rubbing or massaging motion, often repeated and soothing. Use 撫でる for petting an animal or stroking hair; use さする for rubbing someone's back to comfort them.
The English word 'caress' can sound overly formal or romantic in Japanese if translated directly as 愛撫する (aibu suru) in casual contexts. For everyday affectionate touch, 撫でる (naderu) or 優しく触れる (yasashiku fureru, 'touch gently') are more natural.
